Description of Present Research:

The present line of inquiry focuses exclusively on the role of NMDA receptors (NMDARs) in triggering electrographic ethanol withdrawal seizures, a prime cause of morbidity and mortality in chronic ethanol abusers. Using the hippocampal blind slice patch-clamp preparation, recordings of pharmacologically isolated NMDAR-mediated miniature synaptic events (mEPSCs) will be made from organotypic hippocampal tissue cultures (explants) in the setting of chronic ethanol administration, and at various time-points after withdrawal of ethanol. Withdrawal-induced changes in NMDAR-mediated signaling will be temporally correlated with the onset of electrographic seizures detected via field potential monitoring of hippocampal discharge patterns. Confocal imaging of GFP-tagged NMDARs expressed by explanted hippocampal neurons will be employed to directly visualize chronic ethanol-induced NMDAR trafficking, and to monitor time-dependent changes in NMDAR expression at various time points following ethanol withdrawal.
